If screening hits are obtained from the Malaria Box, the following evaluation is suggested to assess the initial quality of the hit:
- Check whether the chemotype is known for the particular target/organism.
- Prioritize the hits based on new information about the drug-like properties. The in vitro metabolism as well as the in vivo pharmacokinetic experiments (single oral dose in mouse) on the Malaria Box compounds are almost complete. MMV will report this data in due course, meanwhile researchers are welcome to contact MMV regarding data availability.
- Confirm activity from a fresh solid sample.
- Check through the original screening data and Malaria Box to see whether there are near neighbors which have activity, and then consult the cytotoxicity data to ensure the biological effect observed is not due to a general cytotoxicity.
There are many commercial suppliers that are able to supply analogues of Malaria Box compounds. These suppliers are listed on searchable websites including emolecules and Chemspider. Alternatively, researchers are welcome to contact MMV to obtain a list of commercially available near neighbors for their hits.
